■背景と問題意識
医薬品や食品などの製造プロセスでは、溶液から目的の固体を回収する「非溶媒晶析」と呼ばれる技術が広く用いられています。このプロセスにて、目的物質が固体として発生せず、溶液中に別の液相が生じて2つの液体に分かれる現象は「オイルアウト(油状分離)」と呼ばれます。
オイルアウトは結晶化を阻害し、製品の品質安定性や再現性を低下させることが多いため、これまでの研究では主にオイルアウトを回避・防止する手法が検討されてきました。しかし、オイルアウト発生時に形成される局所的な溶液組成やその環境を活用した結晶化に関する知見は十分に整理されていませんでした。

“Non-solvent crystallization” is a widely used technique in the manufacturing processes of pharmaceuticals and foods to recover a target solid from a solution. During this process, a phenomenon sometimes occurs where the target substance does not immediately form a solid; instead, a separate liquid phase emerges within the solution, resulting in a separation into two liquid phases. This phenomenon is known as “oil-out” (or oily phase separation).
Since oil-out often hinders crystallization and compromises product quality stability and reproducibility, previous research has primarily focused on methods to avoid or prevent it. However, there has been insufficient understanding regarding crystallization processes that leverage the unique local solution compositions and environments formed during oil-out.
■ Research Approach and Novelty
This study adopts a perspective that actively utilizes oil-out—a phenomenon typically avoided—to systematically investigate how the unique solution compositions generated by oil-out and operational conditions (such as stirring speed and temperature) affect crystal particle characteristics (e.g., primary particle size, secondary particle size, and aggregation behavior).
Impact of unique solution composition: Experimental results demonstrated that the high degree of supersaturation within the droplets formed during oil-out promotes nucleation, thereby influencing the reduction in primary particle size and changes in the number of aggregated particles.
Impact of operational conditions (stirring speed and temperature): It was confirmed that primary particle size decreases as stirring speed increases or temperature drops—changes driven by variations in shear stress and supersaturation levels. Furthermore, the study demonstrated that primary and secondary particle sizes could be varied by approximately twofold through adjustments to stirring speed and temperature.
